Enhanced production of pyrroloquinoline quinone expressed from Escherichia coli via adding signal peptide

Abstract

The signal peptide gene pelb from vector pET-22b was inserted into pET-28a-pqq, resulting in a new vector pET-28a-pqq. Through transformation and screening, a recombinant E.coli BL21(pET-28a-pqq) was acquired to express pyrroloquinoline quinone (PQQ). This strain produced 40.73mg/L of PQQ, which was 24.36% enhancement in comparison with the control strain E.coli BL21(pET-28a-pqq). This result implied the involvement of signal peptide in PQQ secretion or biosynthesis. Besides, both strains showed high PQQ productivity and yield on a biomass basis in the first 3h of fermentation. In particular, E.coli BL21(pET-28a-pqq) demonstrated an increase in PQQ production and active cell growth after 21h, indicating the synergistic relationship between PQQ biosynthesis and biomass.
