Preparation and characterization of 10-hydroxycamptothecin nanoparticles
10-Hydroxycamptothecin (HCPT) nanodispersions have been prepared by using an anti-solvent precipitation method.The influence of different solvents on the nanodispersion of 10-hydroxycamptothecin were investigated,with water as anti-solvent.DMSO was chosen as solvent to prepared nanosized 10-hydroxycamptothecin. The influence of the key parameters,including the volume ratio of solvent and anti-solvent and the weight of PVP,on the particle morphology,size and size distribution have been explored.The average particle size was about 95nm.The XRD spectra show that compared with the crude HCPT,the nano HCPT has lower crystallinity,and the FT-IR spectra show that the nano HCPT and crude HCPT have the same chemical structure and composition.The dissolution rate of nano 10-hydroxycamptothecin was significantly faster than that of the raw material.In the first 30min,the cumulative dissolved amount of the nano 10-hydroxycamptothecin was nearly 100%,while the dissolution of the raw material was only 11.6%.
